In The Big Bus volgen we het 75 ton zware en 32 wielen tellende monster tijdens zijn eerste rit van New York naar Denver. De chauffeur van de magistrale bus is zelf al even meesterlijk. Zijn bijrijder heeft nogal eens last van een black-out en rijdt liever in de berm dan op de weg. Een aantal kwaadgezinde lieden heeft het van meet af aan op de bus gemunt, wat tot de nodige hilarische en spannende taferelen leidt.
